1.  Experiments:  Mitochondrial Respiration and Photosynthesis
     in Isolated Thylakoids

In this month's newsletter, two experiments that utilize a dissolved oxygen electrode and a cellular respiration/photosynthesis chamber are featured. 

In the mitochondrial respiration experiment, the amount and rate of oxygen consumed by mitochondria in the presence of different combinations of substrates, inhibitors, and uncouplers indicates the flow of electrons through the molecules that are part of the electron transport chain (ETC).  The energy released as electrons pass through the ETC is used to synthesize ATP. 

In the photosynthesis experiment, the amount and rate of oxygen produced by chloroplasts indicates the light-driven flow of electrons through the photosystems that convert light energy to chemical energy.  The energy captured is used to synthesize sugars from carbon dioxide and water.

In these experiments, the students use  the ISE-730 dissolved oxygen electrode, the DO2-100 current to voltage adapter that converts the output of the ISE-730 to a measurable voltage, and the RPC-100 respiration/photosynthesis chamber.

2.  Articles:  The RPC-100 Respiration/Photosynthesis Chamber,
    the ISE-730 Oxygen Electrode, and the DO2-100 Current to
    Voltage Adapter.

The RPC-100 is a reaction chamber designed for studying cellular respiration or photosynthesis.  The chamber is filled with a mixture of cells or cell fractions, buffers, and reagents that react to either consume or produce oxygen.  The amounts and rates of oxygen, consumed or produced, are measured using an ISE-730 oxygen electrode placed in the chamber.  The current output of the oxygen electrode is converted to a voltage output by the DO2-100 adapter which also amplifies the output voltage before it is recorded.

5.  LabsOnCD - A Requirement for Distance Learning
     Physiology Curriculum
